Description:

Purpose

This App can be used to fit a standard curve like Concentration vs Absorbance. Such a curve can be used to determine concentrations in "unknown" samples with their measured absorbance.

This method is commonly used to analyze RIA or ELISA Data.

Installation

Download the file "stdCurveAnl.opx", and then drag-and-drop onto the Origin workspace. An icon will appear in the Apps gallery window.

Operation

Click the App icon to open a toolbar.

  • Step 1: Create new worksheet to input data
    1. Click button 1 to open the dialog.
    2. Select Standard Curve Type. If you have replicate standard curve data, choose Single X and Replicate Y. The App performs concatenate fitting for replicate data.
    3. Enter labels for X column and Y column.
    4. Click OK to create a new worksheet. It contains 2 regions highlighted with different colors.
    5. Enter standard curve data in the 1st region. Enter unknown sample data in the 2nd region.

  • Step 2: Perform standard curve analysis
    1. Activate the worksheet created in Step 1 and click button 2 to open the dialog.
    2. In the Input tab, the ranges of standard curve data and sample data are automatically selected. 
    3. In the Settings tab, select a fitting function to fit a standard curve. Make changes to parameters including initial values, fix parameters, lower and upper bounds, etc.
    4. Click Preview button to display fit curve and interpolated data points.
    5. In the Fit Control tab, change Max. Number of Iterations and Tolerance if fitting fails to converge.
    6. In the Quantities and Plots tab, choose quantities and plots to output in the report.
    7. Click OK button to output report sheets.

  • Step 3: Create an HTML report

    Keep the report sheet active and click button 3. An HTML report will be created, and then you can export it to webpage.

Sample OPJU File
This app provides a sample OPJU file.  Right click the App icon in the Apps Gallery window, and choose Show Samples Folder from the short-cut menu. A folder will open. Drag-and-drop the project file StandardCurveAnalysisSample.opju from the folder onto Origin. The Notes window in the project shows detailed steps.
Note: If you wish to save the OPJU after changing, it is recommended that you save to a different folder location (e.g. User Files Folder).
